چکیده

Both human epidermal growth factor receptor 2 (HER-2/neu) and VEGF overexpression correlate with aggressive phenotypes and decreased survival among breast cancer patients. Concordantly, the combination of trastuzumab (anti-HER2) with bevacizumab (anti-VEGF) has shown promising results in preclinical xenograft studies and in clinical trials. However, despite the known antiangiogenic mechanism of anti-VEGF antibodies, relatively little is known about their effects on the pharmacokinetics and tissue distribution of other antibodies. This study aimed to measure the disposition properties, with a particular emphasis on tumor uptake, of trastuzumab in the presence or absence of anti-VEGF. Radiolabeled trastuzumabwas administered alone or in combination with an anti-VEGF antibody to mice bearing HER2-expressing KPL-4 breast cancer xenografts. Biodistribution, autoradiography, and single-photon emission computed tomography–X-ray computed tomography imaging all showed that anti-VEGF administration reduced accumulation of trastuzumab in tumors despite comparable blood exposures and similar distributions in most other tissues. A similar trend was also observed for an isotype-matched IgG with no affinity for HER2, showing reduced vascular permeability to macromolecules. Reduced tumor blood flow (P < 0.05) was observed following antiVEGF treatment, with no significant differences in the other physiologic parameters measured despite immunohistochemical evidence of reduced vascular density. In conclusion, anti-VEGF preadministration decreased tumor uptake of trastuzumab, and this phenomenon was mechanistically attributed to reduced vascular permeability and blood perfusion. These findings may ultimately help inform dosing strategies to achieve improved clinical outcomes. Mol Cancer Ther; 11(3); 752–62. 2012 AACR.

Anti-Angiogenic and Anti-Proliferative Effects of Physalis Alkekengi Hydroalcholic Extract on Breast Cancer in Mice

Background & Objective: The Physalis alkekengi is a herbaceous plant belonging to Solanaceae family. According to research, this plant can help inhibit cancer cell growth. Materials & Methods: In this study, the extract of Physalis alkekengi was used to suppress angiogenesis in Balb/c mice. First, the MC4L2 cells were injected in the mice breastbone and the tumor began to grow in this area, th...

Anti-cancer effects of the combined treatment of trastuzumab and decoy oligodeoxynucleotides to target STAT3 transcription factor on SK-BR-3 breast cancer cell line

Introduction: Breast cancer is the most common malignancy in the female population and is the leading cause of death. Surgery, chemotherapy, radiotherapy, and monoclonal antibody (trastuzumab) therapy are common and standard treatments for this cancer. However, there are significant limitations in the treatment of this disease by using regular methods. Given the role of transcription factors (T...

Pharmacokinetics and Biodistribution of (86)Y-Trastuzumab for (90)Y dosimetry in an ovarian carcinoma model: correlative MicroPET and MRI.

UNLABELLED Preclinical biodistribution and pharmacokinetics of investigational radiopharmaceuticals are typically obtained by longitudinal animal studies. These have required the sacrifice of multiple animals at each time point. Advances in small-animal imaging have made it possible to evaluate the biodistribution of radiopharmaceuticals across time in individual animals, in vivo. MicroPET and ...

Gene Expression Changes in Pomegranate Peel Extract-Treated Triple-Negative Breast Cancer Cells

Background: Triple-negative breast cancer (TNBC) is treated with highly aggressive non-targeted chemotherapies. Safer and more effective therapeutic approaches than those currently in use are needed. Natural pomegranate peel extract (PPE) has recently been found to inhibit breast cancer progression; however, its mechanisms of action remain unclear. We hypothesized that transcriptional chan...
